Abstract
The domain organization of a sigma70 subunit fragment from the RNA polymerase of Escherichia coli is determined using limited proteolysis, N-terminal sequencing and mass spectrometry. The fragment's crystal structure reveals an entirely helical, V-shaped protein. The structure indicates that DNA interactions may be inhibited in the absence of RNA polymerase. Moreover, it can be used to interpret a large number of genetic and biochemical analyses.
