Abstract
The Taxonomy of organisms has long been a point of interest for scientists. The availability of DNA sequencing has made this field even more prominent. However, there are still improvements that can be made to this method. One such improvement is to use barcoding of the rRNA (rrn) Operon in order to extract the genetic information from these species and possibly from metagenomes as well. A database of over 1,000 microeukaryotic genomes containing various supergroups was created, allowing for the rrn Operons to be extracted and assembled. Then, using a tree-building program the taxonomies of these species were compiled into a phylogenetic tree which was deemed sufficiently accurate after comparing it with the 18S trees of Hug et al 2016 and multigene tree of Strassert, et al, 2019. For more evidence, assembled metagenomes were then processed using the same methods and added to the tree. This tree was deemed sufficiently accurate, using bootstrap values as a comparison to the previously compiled tree. With this in mind, it can be shown that using the rRNA Operon as a barcode has shown potential to be both taxonomically correct and an improvement from 18S with the hope of providing taxonomic consistency and future discoveries.
